Leptin (22–56) is a synthetic peptide fragment corresponding to amino acid residues 22 through 56 of the full-length leptin protein. Leptin is a cytokine-like signaling molecule involved in the regulation of energy balance, metabolic signaling, and neuroendocrine communication through its interaction with the leptin receptor (Ob-R), a member of the class I cytokine receptor family. [1]

Mechanism of Action

Leptin is involved in the regulation of metabolic signaling and energy-related processes through its action on hypothalamic pathways. It functions as a signaling molecule between adipose tissue and the central nervous system, helping coordinate physiological energy balance mechanisms. [1]

Leptin also contributes to reproductive axis signaling, acting as a permissive factor for activation of the hypothalamic–pituitary–gonadal (HPG) axis. It influences gonadotropin-releasing hormone (GnRH) secretion and supports normal endocrine signaling associated with reproductive function. [2]

In addition, leptin has been shown to influence skeletal development pathways and growth-related signaling centers, linking metabolic status with structural and developmental processes. These effects highlight the interaction between endocrine signaling and skeletal system regulation. [4]

Within the central nervous system, leptin is involved in neuronal signaling and memory-associated pathways, particularly through its activity in the hippocampus. It has been shown to modulate synaptic plasticity and neuronal communication, which are essential processes in cognitive research models. [2]

In transgenic mouse models of neurodegenerative research, leptin has been observed to influence memory-related performance and reduce pathological markers, including amyloid-related accumulation. These effects are associated with modulation of intracellular signaling pathways such as JAK2/STAT3 and improvements in neuronal signaling activity. [3]
