Research Findings on Pregnanolone

Anti‑inflammatory and Immune Modulation Effects (Preclinical Research)

In laboratory studies using cultured immune cells, pregnenolone has been shown to influence innate immune signaling pathways at the molecular level. Specifically, pregnenolone was found to promote the targeted degradation of key signaling proteins involved in toll‑like receptor (TLR) pathways, such as TIRAP and TLR2, which play major roles in activating inflammatory responses. This action resulted in reduced secretion of pro‑inflammatory cytokines like tumor necrosis factor‑α (TNFα) and interleukin‑6 (IL‑6). This suggests that pregnenolone can modulate inflammatory signaling in macrophage and microglial cell models studied in vitro. 

Neuroactive Steroid Properties in Research Models

In laboratory research, analogs of pregnanolone (a neuroactive form of pregnenolone) have shown effects on nervous system function. For example, studies in developing animal brains demonstrated that pregnanolone glutamate, a derivative compound, can interact with key neurotransmitter receptor systems, such as GABA and NMDA receptors. It also exhibited strong age‑related anticonvulsant effects in seizure models.
